Molecular and Cellular Pathobiology p38a Inhibits Liver Fibrogenesis and Consequent Hepatocarcinogenesis by Curtailing Accumulation of Reactive Oxygen Species

چکیده
Most hepatocellular carcinomas (HCC) develop in the context of severe liver fibrosis and cirrhosis caused by chronic liver inflammation, which also results in accumulation of reactive oxygen species (ROS). In this study, we examined whether the stress-activated protein kinase p38a (Mapk14) controls ROS metabolism and development of fibrosis and cancer in mice given thioacetamide to induce chronic liver injury. Liver-specific p38a ablation was found to enhance ROS accumulation, which appears to be exerted through the reduced expression of antioxidant protein HSP25 (Hspb1), a mouse homolog of HSP27. Its reexpression in p38adeficient liver prevents ROS accumulation and thioacetamide-induced fibrosis. p38a deficiency increased expression of SOX2, a marker for cancer stem cells and the liver oncoproteins c-Jun (Jun) and Gankyrin (Psmd10) and led to enhanced thioacetamide-induced hepatocarcinogenesis. The upregulation of SOX2 and c-Jun was prevented by administration of the antioxidant butylated hydroxyanisole. Intriguingly, the risk of human HCC recurrence is positively correlated with ROS accumulation in liver. Thus, p38a and its target HSP25/HSP27 appear to play a conserved and critical hepatoprotective function by curtailing ROS accumulation in liver parenchymal cells engaged in oxidative metabolism of exogenous chemicals. Augmented oxidative stress of liver parenchymal cells may explain the close relationship between liver fibrosis and hepatocarcinogenesis. Cancer Res; 73(1); 1–10. 2012 AACR.

Effect of Hesperetin on the level of reactive oxygen species (ROS) in gastric cancer stem cells: Short Communication
Intracellular reactive oxygen species (ROS) play an important role in cancer stem cell (CSC) function. Hesperetin (Hst) is a flavonoid that has been shown to affect cellular ROS level. The goal of this study was to investigate the effect of Hst on the level of ROS in gastric CSCs (GCSCs). MTT assay was used to evaluate cell survival. Cellular ROS level was measured using 2′,7′-dichlorofluoresci...

Nonalcoholic fatty liver disease (NAFLD) is one of the most important chronic liver diseases worldwide and has garnered increasing attention in recent decades. NAFLD is characterized by a wide range of liver changes, from simple steatosis to nonalcoholic steatohepatitis, cirrhosis, and hepatocellular carcinoma. The blurred pathogenesis of NAFLD is very complicated and involves lipid accumulatio...
متن کاملL-carnitine inhibits hepatocarcinogenesis via protection of mitochondria.
Hepatocellular carcinoma is usually preceded by chronic inflammation. However, the molecular mechanism in hepatocarcinogenesis is not well known. Recently, we reported that mitochondrial dysfunction plays an important role in hepatocarcinogenesis via the production of free radicals. Furthermore, we proved that L-carnitine effectively protects mitochondrial function in vivo. Therefore, we invest...
متن کاملObesity, diabetes mellitus, and liver fibrosis.
Obesity is a global epidemic with more than 1 billion overweight adults and at least 300 million obese patients worldwide. Diabetes is characterized by a defect in insulin secretion or a decrease in sensitivity to insulin, which results in elevated fasting blood glucose. Both obesity and elevated fasting glucose are risk factors for nonalcoholic fatty liver disease, a disease spectrum that incl...
متن کاملMolecular and Cellular Mechanisms of Chemically Induced Hepatocarcinogenesis
Liver is the main organ of xenobiotic biotransformation. Since biotransformation may generate highly mutagenic active metabolites and reactive oxygen species, liver cells are both the producers and targets for these compounds. Xenobiotics can lead to liver tumour formation via genotoxic or non-genotoxic mechanisms. Non-genotoxic chemicals are often inducers of monooxygenase reactions depending ...
